Pengaruh Ekstrak Akar Eurycoma longifolia Jack (pasak bumi) terhadap Peningkatan Kadar Testosteron, LH dan FSH pada Tikus Jantan Sprague Dawley

Ekstrak Eurycoma long Jack atau Pasak Bumi (Pas), secara tradisional telah lama digunakan untuk meningkatkan atau mengembalikan potensi sekxual pria. Penelitian ini dirancang untuk medapatkan bukti ilmiah dengan melibatkan 30 ekor tikus jantan Sprague Dawley (umur 90 han) sebagai binatang coba . Penelitian ini menggunakan rancangan Post test only control Group Design. Binatang coba dikelompokkan menjadi 3 kefompok (inasing-masing 10 tikus), terdiri dan kelompok kontrol (K) menerima 1 ml aquades. Sedang keloinpok perlakuan 1 (P-1) menerima l ml ekstrak Pas (25 mg) dan kelompok perlakuan 2 (P-2) menerima 2 ml ekstrak Pas (50 mg). Untuk adaptasi tikus dimasukkan kandang secara individual selama satu minggu, selanjutnya setiap jam 7 pagi tikus mendapatkan perlakuan secara oral dengan menggunakan acu firm blunt type needle selama 7 hari . Pada akhir perlakuan, sampel darah diambil dengan mengaspirasi sinus orbitalis. Kadar LH dan ESH diperiksa den gan Immuno Radiometric Assay (IRMA), sedangkan kadar Testosteron diperiksa dengan Radio Immuno Assay (RIA).
 
Hasil peneliian ini menunjukkan bahwa ekstrak Pas 2 ml meningkatkan kadar LH secara bermakna (P=0.04), kadar ESH (P=I). 009) demikian pula dengan kadar Tesiosteron (P =0. 031) dibanding kontrol. Sementara itu 1 ml Pas hanya meningkatkan kadar Testosteron secara bermakna (P=0.035}, tetapi tidak meningkatkan kadar LH (P=0.07/) maupun FSH (P=0.059) dibanding kontrol.

Dideduksi dan hitungan matematik pada korelasi kadar LH dengan peningkatan kadar Testosteron dapat disimpulkan bahwa peningkatan kadar Testosteron disebabkan oleh efek tidak langsung melalui peningkatan LH dan efek fangs ung melalui konversi sterol dalam Pas menjadi Testosteron dijaringan perifer. Sebagai simpulan, penelitian ini memberikan bukti ilmiah pendahuluan mengenai penggunaan Pas untuk meningkatkan atau mengembalikan potensi seksual pria.

Traditionally, Eutycoma longifolia Jack or Pasak Bumi (Pas) extract have been using to enhance or restore male sexual potencies. This particular study had designed to gain scientific consent involving 30 male of Sprague Dawley rats (90 days old) as animal model. Post-test only control group design was adopted. The animal models were divided into 3 group (10 rats each) i.e. K group received] ml of water as control group and treated group designated as P-I received 1 ml of Pas extract (25 mg/mI) and P-2 received 2 ml of Pas extract respectively. As adaptation protocol, these rats were individually caged for one week then treated orally using Acufirm blunt type needle, every morning at 07.00 a. m., for 7 days. At the end of treatment, blood samples were obtained by orbital sinus aspiration. The concentration of LH and FSH were assessed by Immuno Radiometric Assay (IRMA), while testosterone concentration was assessed by Radio Immuno Assay (RIA).
 
The result of this study showed that 2 ml Pas extract (P-2 group) significantly induce increasing LH concentration (P=0.04),ESH concentration (P=0. 009), as well as testosterone concentration (P=0. 031) compared to those of control group (K). However, 1 ml Pas (P-i group) only significant induce increasing testosterone level (P=0.035), but not for LH (P=0.071) nor FSH (P=0.059) compared to those of control group.
 
Deducted from mathematical equation on correlation of LH concentration and Testosterone level achievement, it could be concluded that increasing Testosterone level was due to indirect effect of LH improvement and direct effect of Pas conversion (sterol conversion) to Testosterone on the peripheral tissues. As a conclusion, this study apparently provide scientific preliminary approval of traditional believe of using Pas to enhance or restore male sexual potencies.
