Perbedaan Diet Ekstrak Rumput Laut (Eucheuma Sp) dan Insulin Dalam Menurunkan Kadar Glukosa Darah Tikus Putih (Rattus Norvegicus) Hiperglikemia

Background: Eucheuma Sp (Eu sp) is one of red seawes (Rhodophyceae ) taht has beeb proven to reduce hyperglicemia in rat. The aim of this study was to know thediffernt ability of seawed extract ((Eu sp) and insulin in reducing blood glukose level of hyperglikemiarats.
Methods : The randomized pre and post test control group was conducted in rats (Rattus norvegicus) in aniaml experiment Unit in Gajah Mada University . tenty four
Rattus norvegicus , were divided into 2 groups. Group 1 was treated with standart diet +1 /kgBW Eu sp ; group II was trated with stndart diet + 1,25 g/kg BW Eu sp ; Group III Was treated with standart diet +1,5 g/kg BW Eu sp ; Group IV was Treated with standart diet + insulin injaction with dose of 3,78 UI /kg BW. The Rattus norvegicus were made hyperglcemic by aloksan injection with dose of 125 mg/kgBW On the 7th day . The blood glucose level was measured before tratment (pre – pandial ) and 2,5 hours aftrr treatment with glucose 1,35 gr orraly (post – prandial) Eu sp extract diet was adsministered every day for 14 days. Blood glucose levels were measured two times within 7 days intervals using Enzymatic Colorimetric Test “ GOD- PAP” method. Statisticanalysis use SPsS for Window 11.00
Result : Adsministration of Eu sp 1;1,25; 1,5 gr/KgBW/day and insulin injection 3.78 UI/Kg BW/Day reduced pre and post prandial blood glucose significantly (Kruskall – wallis, p=0,00) .the highest reduction was observed in insulin groups in which pre prandial blood glucose level was noraml but post prandial level was still high. There werew signficant differences in pre – prandial and post prandial blood glucose between groups (Mann – Whitney P ,2 ,3,4 =0,00)
Conclusion: Reduction of Blood glucose level after Eu sp diets were lower than after given insulin injection

The Different ability of seawed extract (Eucheuma Sp) and Insulin in reducing blood glucose level of hyperglycemic rats

latar Belakang : rumput laut spesies Eucheuma (Eu sp) merupakan salah satu jenis rumput laut merah (Rhodophyceae ) yang telah terbukti dapat menurunkan kadar glukosa darah tikus hiperglikemia. Penelitian ini bertujuan untuk mengetahui perbedaan kemampuan diet ekstrak EU sp dalam menurunkan kadar glukosa darah tikus putih (Rattus norvegicus) hiperglikemia dibandingkan dengan insulin
Metode : Penelitian ini eksperimental dengan desain Randomized Pre and Post – Test controul group , dialakuakn di unit pengembangan hewan percobaaan (UPHP) UGM Y ogyakarat. Dua puluh empat ekor tikus putih jantan (Rattus norvegicus) Dibagi acak menjadi 4 kelompok perlakuan . Kelompok 1 , Diberi diet standart + Eu sp 1g/kgBB/hari
Kelompok II, diberi diet standart + Eu sp 1,25 g/kg/BB/hari. Kelompok III , diberi diet standart + Eu sp 1,5 g/kgBB/hari
Kelompok IV diberi diet standart + injeksi insulin dengan dosis 3,78 UI /kg BB/ hari . semua tikus dibuat hiperglikemiadengan injeksi aloksan dosis 125mg/ kg BB pada hari ke -7 . Pengukuran kadar glukosa darah tikus dengan metode Tes Toleransi glukosa oral (TTGO) . Kadar glukosa darah diamati sebelum perlakuan (pre – prandial ), 2,5 jam sesudah perlakuan dan pemberian beban glukosa 1,35(post – prandial) . Pemberian sediaan uji diberiakn setiap hari selama 14 hari. kadar glukosa darah diukur 2 kali denagn intervel waktu 7 hari menggunakan metode Enzymatic Colorimetric Test “ GOD- PAP” . ANALISA DATA DILAKUKAN DENGAN STATISTIK NON- parametrik menggunakan SPSS for Window 11.00
Hasil: Pemberian Eu sp 1; 1,25 ;1,5 gr/KgBB /hari dan Insulin 3,78UI/KGbb/Hari mamapu menurunkan secara bermakan kadar gula darah tiap kelompok baik pre prandial maupun post – prandial (Kruskall –Walis P 1,2,3,4 = 0,00)
Penurunan tertinggi terajdi pada kelompok yang mendapatkan insulin , dengan kadar gula darah pre- prandial mencapai nilai normal , namun ,masih tinggi pada post –aprandial . Perubahan kdr gula darah pre maupun post – prandial antar kelompok semua secara bermakna (Mann – Whitney P ,2 ,3,4 =0,00)
Simpulan : Penurunan kadar gual darah setelah pemberian berbagai dosis Eu sp lebih rendah daripada injeksi insulin
