Pengaruli Diet Ekstrak Rumput Laut (Eucheuma sp) terhadap Kadar Glukosa Darah Tikus Putih (Raltus norvegicus) Hiperglikemik

Lalar belakang: Eucheuma sp merupakan salah satu jenis dart rumput laut merah (Rhodophyceae) yang mempunyai kandungan karagenan sebagai senyawa serat golongan binding of cation. Penelitian ini bertujuan untuk mengetahui pengaruh Eucheuma sp terhadap kadar glukosa darahpada tikus hiperglikemik
Metoda: Penelitian eksperimental dengan desain Randomized Pre and Post-Test Control Group, dilakukan di Unit Pengembangan Hewan Percobaan (UPHP) UGM Yogyakarta . Duo puluh empat ekor tikus putih jantan (Rattus norvegicus) dibagi acak menjadi 4 kelompok perlakuan. Kelompok I adalah kontrol negatjf diberi diet standart. Kelompok II, diberi diet standart + diet Eucheuma sp dengan dosis I gr/kgBB/hari. Kelompok III, diberi diet standart + diet Eucheuma sp dengan dosis 1,25 gr/kgBB/hari. Kelompok IV, diberi diet standart + diet Eucheuma sp dengan dosis 1,5 gr/kgBB/hari. Tikus dibuat hiperglikemia dengan injelisi aloksan dosis 125 mg/KgBB dan dicapai pada han ke-7. Pengukuran kadar glukosa darah tikus dengan metode Test Toleransi Glukosa Oral (77G0). Kadar glukosa darah diamati saat normal (sebelum diberikan injeksi aloksan), sebelum pemberian sediaan uji (pre-prandial), 2,5 jam sesudah pemberian sediaan uji dan beban glukosa l,35gr (post-prandial). Peinberian sediaan uji diberikan setiap han selama 21 hart. Kadar glukosa darah diukur peniodik dengan interval wa1ctz 7 han menggunakan metode Enzimatic Colorimetrictest “GOD-PAP”.
Hasil Uji Kruskal-Wallis terhadap di Pre (p =0,00). Uji Kruskal-Wallis terhadap di Post (p=O, 00). Uji Mann- Whitney di Pre kelompok I dengan kelompok II, III dan l (p=O, demikianjuga di Pre kelompok 11 dengan 111 dan IV (p=O,04). Uji Mann-Whitney kelompok III dengan IV (p=0,O’l). Uji Mann-Whiyney dt Post kelompok I dengan II, III dan IV (p =0,04), deniikianjuga dt Post kelompok II dengan III dan IV (p =0,0’!). Uji Mann- Whitney dt Post kelompok III dengan IV (p =0, 04).

Background: Eucheuma sp is one of red seaweeds (Rhodophyceae) that has carrageenan as binding kation fiber. The ai of this study is to know of the effect Euheuma sp extract dietary on hyperglycemic rat blood glucose level.
Method: The randomized pre and post test control group was conducted in rat (Rattus norvegicus) in UPHP UGM Yogyakarta. The object of study was 24 Rattus norvegicus, divided into 4 groups. Group I treated with standart diet; Group 11 treated with lgr/KgBW Eucheutna sp extract dietary; Group III treated with 1,25 gr/KgBW Eucheuma sp extract dietaiy; Group IV treated with 1,5 gr/KgBW Euheuma sp extract dietary. The Rattus norvegicus were made hyperglycemic condition by aloxan injection with dose of 125 mg/KgBW at the day seventh. The rat blood glucose level were measured using method glucose tolerance test. The rat blood glucose level were measured at normal condition (before aloxan injection), before Eucheuma sp extract diet (pre-prandial) , 2,5 hours after Eucheuma sp extract diet and orally glucose 1,35gr (post-prandial). Eucheuma sp extract diet were administered every day for 21 days. Blood glucose levels were measured periodically with 7 days intervals using Enzimatic Colorimetrictest “GOD-PAP method.
Result: Kruskal- Wallis test for dt Pre (p=0, 00). Kruskal- Wallis test for dt Post (p—O, 00). Mann- Whitney test for dt Pre between group 1 and ii or 111, 1V (p=zO,O Mann-Whitney test for dt Pre between group 11 and 111 or IV ( p—0,O4). Conclusion: The result showed that all Eucheuma sp extract diet on the control were able to reduce blood glucose leveL Reduction of blood glucose level after Eucheuma sp extract diet with dose 1, Sgr/KgBB was higher than the dose of J,25gr/KgBB and much higher than the dose of I gr/KgBB.
