Pengaruh Pemberian Epigallocatechin Gallate (EGCg) Teh Hijau terhadap Kadar Eosinofil pada Mencit yang Disensitisasi dengan Ovalbumin

Latar belakang: Epigallocatechin gallate (EGCg) inerupakanjenis catechin yang paling besar jumlahnya di dalam daun teh hyau dan memiliki efek menguntungkan bagi kesehatan antara lain sebagai anti tumor; anti mikroba dan anti alergi. Walaupun EGCg dilaporkan mempunyaiperan anti alergi namun efeknya terhadap jumlah sel eosinofil darah belum dilaporkan . Tujuan penelitian ini adalah  untuk melihat pengaruh Epigallocatechin gallate teh hjjau lerhadap kadar eosinofil mencit BALBIc yang disensitisasi dengan oval bumin.
Metoda: Rancangan “post test only group design” dipilih dengan obyek penelitian adalah mencit galur BALBIc, umur 6 minggu sebanyak 24 ekor; yang dibagi menjadi 4 kelompok .Perlakuan berupa pemberian ovalbu”ün EGCg 0,5 mg/hari dan 2 mg/hari. Untuk menginduksi reaksi alergi mencit disensitisasi 100 ug ovalbumin (OVA) dalarn 0,1 rnl  alum adjuvant i.ppada hari  1 dan 14, dan diberi suplementasi EGCg selama 28 hari sebagai imunodulator Pada hari ke 29 diambil sampel dan darah tepi dan limpa untuk dilakukan pemeriksaan . eosinofil / 100 lekosit.
Hasil.: Pada kelompok kontrol, F,, P dan P didapat kan jumlah eosinofil berturut-tunut sebanyak 1,00 ± (0,63); 3,7 (± 0,82); 2,3 ± (0,52) dan 1,3 (± 0,82)dan penurunan ierjadi secara bermakna (p,,=O, 048, p, 000)
Simpulan: Pernberian EGCg mampu menurunkan secara bermakna jumlah eosinofil /100 leukosit darah tepi mencit BALB/c yang disensitisasi OVA.

Background: Epigallocatechin gallale (EGCg is a major form of tea catechin and has several advantage on health, acting as anti tumor; anti microbes and anti allergy. T-lymphocytes have been implicated in the pat hogenesis of allergic disease. Although EGCg is known to have anti allergy role, its effect on the number of eosinophils has not been reported. The objective of this study was to investigate the effect ofEpigallocatechingallatefrom green tea on eosinop hi/s in i9A LB/c mice sensitized with ovalbumin.
Method. The design was a true experimental with post test only group design. Subject were 24 BALB/c mice of 6 week old, devided into 4 groups. The treatments were ovalbumin and EGCg of 0.5 mg/day, and2 mg/day. For the induction of allergic reaction, BAL mice were imunized intraperitoneally with /00 ug ova/bumin (OVA) in 0.1 rn/alum on day I and day 14. BALB/c mice were given EGCg sup/ementation for 28 days as imunornodulator agent. Peripheral blood samples and spleenocytes were taken on day 29. Eosinophi/s 100 leukocyte were counted by dj/jerential counting of leukocyte from peripheral blood smear staining with Giemsa.
Results: The mean of eosinophil in control group was 1.00 ± 0.63 and in the groups treated with ovalbumin, but without EGC’g (P) was 3.7 ± 0.82; in P OVA (+), EGC’g 0.5 mg/day/eosinophils was difference was sign (P,,=O. 048; P,,0. 000)2.3 ± 0.52 and in P OVA (+), EGCg 2.0 mg/day! eosinophils was 1.3 ± 0.82.
Conclulion: This study revealed that EGCg could decrease the number eosinophils/I00 leukoc.ytes in peripheral blood significantly.
