Pengaruh diet minyak ikan kaya Omega -3 (Ώ-3)pada produksi interferon gamma makrofag dan survival Mencit Balb/c ang mendapat vaksinasi BCG

Latar Belakang: Manfaat minyak ikan kaya Ώ -3 terhadap penyakit kardiovaskuler dan inflamasi /autoium sudah tidak diragukan lagi . Tetapi penelitian terakhir menunujukkan bahwa penggunaan minyak ikan kaya Ώ -3 dalm jangka waktu tertentu dapat menurunkan imunitas seluler . Disisi lain ada imunostimulaltor yaitu BCG Yang dapat meningkatkan respon imunitas seluler type I. Study ini bertujuan membuktikan efek diet minyak ikan kaya Ώ -3 dan upaya vaksinasi BCG terhadap produksi interferon gama (IFN-ץ makrofag dan survival mencit terhadap infeksi

Metode :
72 ekor mencit jantan tua strain BALB/c dibagi menjadi 2 bagian untuk penelitian survival (48 ekor) dan laboratoris (2 ekor) . Masing masing bagian baik untuk penelitian survival maupun laboratoris terdiri dari :4 kelompok yang semuanya mendapatkan makanan standart. Pada kelompok kontrol (k) , mecit tidak mendapatkan perlakuan , sedangkan kelompok BCG (BCG) divaksinasi dengan 0,1 cc BCG intra peritonial pada hari ke 14 dan ke 24, Kelompok minyak ikan (MI) mendapat diet 5% minyak ikan kaya Ώ -3   , sedangkan kelompok minyak ikan+ BCG (MB) mendapatkan perlakuan kombinasi BCG dan MI . Pada hari ke 28, mencit untuk penelitian laboratoris disuntik secara intervena dengan 104 Listeria monocytogeneses hidup . sedangkan yang diamati untuk survival disuntik dengan 106 kuman yang sama . Mencit dalam penalitian laboratoris dibunuh pada hari ke 33 , sedangkan pada kelompok survival terus diamati sampai 28 hari setiap 12 jam. Perbedaan produksi IFN-ץ makrofag disnslisis dengan Kruskal – Walis test yang dilanjutkan Mann-Wwhitney U. Survival dianalisis dengan metode Kaplan –meier dan log rank statistic

Hasil:
Produksi IFN-ץ makrofag pada kelompok MI lebih rendah daripada MB (p=0,026) , Survival pada kelompok MB paling tinggi tetapi tidak didapptakan pebedaan yang bermakna antar kelompok perlakuan (p=0,085)

Simpulan :
BCG dapat meningkatkan produksi IFN-ץ amkrofag yang tertekan akibat penggunaan minyak ikan kaya Ώ -3 . Pada penelitian survival tidak didapatkan perbedaan kemampuan bertahan hidup antar kelompok percobaan

The Influence of rich Ώ -3 Oil diet on Macrophages Gamma Interferon Production and survival of Balb/c Mice Received BCG.

Background: It is known that  rich Ώ -3 Oil gives benefict effect on helth especially in cardiovascular and inflamantory autoimune disease. Recent study , however , mentioned an impaiment of cellular immunity when fish oil is consumed for certain period. On the other hand, BCG vaccination is known as an immunostimulator through increasing celular immunity via type I response. The study was to see the effect rich Ώ -3 fish oil diet and the use of bcg vaccination in machropage interferon   IFN-ץ  produc tion and survival againts infection  

Methods:
The 72 male old BALB/c mice were divided into survival group (48 mice) and laboratory group (24 mice) . Both survival and laboratory group were then divided into four group and received standart lab diet daily.the control group (c) received no other additional tretment , while the BCG group (BCG) received intra-peritoneal injection of 0,1 cc BCG at day 14 and 24 . The fish oil group (FO) received 5% rich Ώ -3 Oil and the fish oil + BCG (FB) received combination tratment of BCG and FO Groups . Mice were intravenously injected with 104 (laboatory group) and 106 (survival group) alive L. monocytogeneses at days 28th .Mice for laboratory study were sacrified at day 33 , While in survival study they were followed up every 12 hours for 28 days. The macrophages IFN-ץ production were analyzed by Kruskall Wallis followed by Mann Whitney U test. Survivals were analyzed by Kaplan –Meier and log rank statistics

Result:
The macrophages IFN-ץ production in group was significantly lower than FB group (p=0,026). Survival in FB group was highest but there were no signifficant differences within the group (p=0.085).

Conclusion : Rich Ώ -3  fish Oil has a suppresive effect in macrophages IFN-ץ production while additional treatment with BCG was able to restore it . Although the survival in FB group was highest, there were no signifficant differences within group.
