Kerusakan Sel-sel Reproduksi dan Penurunan Jumlah Anak Tikus Putih Jantan Galur Swiss Akibat Paparan Timah Hitam Per Oral

Latar belakang: Timah hitam (Pb,) ,nerupakan suatu hahan pencemar lingkungan yang mampu menimbulkan kerusakan sistem cacat jantung, ginjal, reproduksi, respirasi dan endoknin. Kerusakan sistem reproduksi akan memnpengaruhi jumlah anak yang dilahirkan, Tujuan penelitian ini untuk menganalisis kerusakan sel sel reproduksi dan penurunan jumlah anak tikus putih galur Swiss setelah, pemberian berbagai konsentrasi larutan Ph-nitrat.

Bahan dan cara kerja
: penelitian eksperimenial lahoratonium ini adalah post-test randomized control group . Tigapuluh tiga ekor tikus jantan galur Swiss berumur 8—10 minggu. dengan berat badan normal, diperoleh dan Laboratorium Biologi Institut Teknologi Bandung secara acak dibagi dalam 3 kelonipok. Tiap tikus dalam kelompok pertama diberi Ph-n itrat yang dilaruikan dalam akuades dengan konsentrasi 800 ppm, sedangkan kelompok kedua diberi Ph-nitrat dengan konsentrasi 400 ppm. Kelompok ketiga sehagai kontrol diheri 0 ppm. Pemberian larutan Ph-nitrat tersebut dilakukan setiap pagi dengan cara dicekok menggunakan semprit 1 ml selama 10 minggu. Pada akhir minggu ke- lO, tikus jantan tersebut dikawinkan dengan tikus betina yang fertil tikus jantan kemudian didekapitasi, dilakukan analisis kualitas sperma dan sediaan histopatologi jaringan testes dianalisis kerusakan sel – sel reproduksi.. Sebelum didekapitasi, tiap kelompok diambil sekelompok tikus untuk diperiksa kandungan Ph—nitratnva pada jaringan testis dengan metode atomic absorben spectrophotameter Dievaluasi pula perbedaan jumlah anak tikus sekelahiran dari tikus betina
 
Hasil : Pembenian larutan Pb—nitrat 800 ppm menyebabkan kerusakan sel – sel reproduksi dan menurunkan jumlah anak sekelahiran secara bermakna. sedangkan pemberian larutan Ph-nitrat 400 ppm memyebabkan kerusakan ringan
 
Simpulan: Pemberian larutan pb-nitrat 800 ppm per oral menyebabkankan kerusakan sel-sel reproduksi dan menurunkan jumlah anak sekelahi ran, sedangkan pemberian larutan dengan konsentrasi 400 ppm peroral dan kerusakan ringan.

Background: Black lead or Ph is one of the waste or materials of environmental pollution that cause the damage of nerve, heart, kidne,g reproductive, respiration, and endocrine system. The damage of reproductive system can affect the amount of babies. The off this research is to analyze the damage of reproductive cells and reduction of amount male white-rat babies oiler exposing of different concentration of Ph nitrate solutions.

Design: The design of this experimental research was post test randomized control group design. 7 of 8—10 weeks healthy male rats strain Swiss with normal body weight obtained from the Biology Laboratory of Bandung Institute of Technology were divided into 3 groups. Pb nitrate dissolved in aquadest in different concentration (800 ppm, 400 ppm and 0 ppm) were distributive to each groups by disonde using I ml spuit every morning for 10 weeks, where as 0 ppm of Ph nitrate was as a control group. the end of tenth weeks, the male rats were copulated with fertile female rats. By using AAS methods one of each rat.groups were sampled to evaluate the contents of Ph in testes before decapitated. The experimental animals were decapitated, testis were prepared histopalhological evaluation such as the damage of reproductive cells and the quality of sperm were evaluated. The amount of rat babies to the sonic birth of fertile female rats were counted.
 
Results: The distribution of Pb nitrate solution of 800 ppm concentration was damage the reproductive cells and caused the reduction of the rat babies in the sonic birth ( The distribution of Pb nitrate solution of 400 ppm concentration caused a slight damage of the reproductive cells and reduction of the rat babies in the same birth.
 
conclusion: Pb-nitrate solution of 800 ppm distributed per oral damnage the reproductive cells and caused reduction of amount rats babies, while the distribution of 400 ppm solution had a slight effect compared with control group. Pb—nitrate, white—rats male strain Swiss, the damage of reproductive cells, reduction of amount rat babies.
